葛根枳椇子洛神花本草酵素的解酒护肝作用研究
以葛根、枳椇子、洛神花为原料，通过益生菌发酵制备本草酵素。筛选菌种比例，分析酵素营养成分，并建立小鼠醉酒模型，考察小鼠醉酒后的醒酒时间、血液乙醇浓度、肝组织抗氧化酶活性含量的变化，探究酵素解酒护肝作用。结果表明，植物乳植杆菌（Lactobacillus plantarum）、罗伊氏粘液乳杆菌（Lactobacillus reuteri）、鼠李糖乳酪杆菌（Lactobacillus rhamnosus）以5∶4∶1比例进行发酵，本草酵素中总酸、粗多糖、游离氨基酸、γ-氨基丁酸、多酚、总黄酮含量分别为21.29 g/kg、12.95 mg/g、3.61 mg/g、86.66 μg/g、4.32 mg/g、0.06 mg/g。酵素实验组小鼠醉酒时间和醒酒时间明显缩短；酒后3 h内小鼠血液乙醇浓度显著降低（P＜0.05）；对比模型组和未发酵提取液组，中、高剂量组小鼠肝组织匀浆中乙醇脱氢酶、超氧化物歧化酶、谷胱甘肽水平均显著升高，丙二醛含量显著降低（P＜0.05），表明葛根枳椇子洛神花本草酵素具有解酒护肝功能。
Abstract
Using Pueraria lobata, Semen Hoveniae and Hibiscus sabdariffa as raw materials to prepare herbal Jiaosu by probiotic fermentation. The bacteria proportion was screened, the nutritional components of ferment were analyzed, and the drunken mice model was established. The changes of sobriety time, blood alcohol concentration and antioxidant enzyme activity content in liver tissues of mice after drunkenness were investigated, and the effect of Jiaosu on anti-alcohol and protecting liver was explored. The results showed that the fermentation ratio of Lactobacillus plantarum, Lactobacillus reuteri and Lactobacillus rhamnosus was 5∶4∶1. The contents of total acid, crude polysaccharide, free amino acid, γ-aminobutyric acid, polyphenol and total flavone in herbal ferment were 21.29 g/kg, 12.95 mg/g, 3.61 mg/g, 86.66 μg/g, 4.32 mg/g, and 0.06 mg/g, respectively. The drunkenness and sobriety time of mice in the ferment group shortened obviously. The blood alcohol concentration of mice significantly decreased within 3 h after drinking (P＜0.05). Compared with the model group and unfermented group, the levels of alcohol dehydrogenase, superoxide dismutase, glutathione in liver tissue homogenate of mice in medium and high dose groups significantly increased (P＜0.05) while malondialdehyde contents decreased, which indicated that the P. lobata, S. Hoveniae and H. sabdariffa herbal Jiaosu had function of anti-alcohol and liver protection.
