油用牡丹“凤丹白”中FAD2-3基因的表达模式分析 摘 要 牡丹是我国特有的名贵花卉,属于芍药属植物,是新兴的一种木本油料作物,它具有非常突出的一个特点,即“三高一低”。“三高”是指产出高,含油率高,品质高,“一低”是指成本低。油用牡丹除了具有观赏和药用价值以外,还具有很高的油用价值。牡丹籽有非常高的出油率,最高可达37%左右,且多为不饱和脂肪酸,在不饱和脂肪酸中尤其亚麻酸含量比较高。FAD2是ω-6脂肪酸脱氢酶的一种,它是植物中催化多不饱和脂肪酸形成的比较重要的酶,能够催化油酸转向亚油酸,影响多不饱和脂肪酸在植物中的含量和比例。ω-3脂肪酸和ω-6脂肪酸都是多不饱和脂肪酸,由于人体自身不能够合成多不饱和脂肪酸,所以只能从食物中获得,ω-6/ω-3脂肪酸比例不易过高,小于五为宜。 本实验以油用牡丹“凤丹白”为实验材料,分别取开花后十天、二十天、三十天的牡丹种子,采用CTAB-LiCl法提取牡丹种子的总RNA,然后逆转录合成cDNA。利用半定量PCR技术检测了FAD2-3基因在牡丹种子发育不同时期表达的变化,为进一步研究该基因的功能奠定了基础。 关键词:油用牡丹,多不饱和脂肪酸,FAD2,半定量PCRAnalysis?of?the?expression?pattern?of?FAD2-3?gene?in?the?oil?peony?Feng?Dan?Bai ABSTRACT Oil Peony is a unique flower in our China, but also a new kind of woody oil crops, with the highlight of the three high and one low characteristics, namely high-yield, high oil content, high quality, low cost. Oil Peony addition to ornamental and medicinal value. As the peony seed oil rate is very high,to reached 24.12% -37.83%, the value of oil is also very high. FAD2 is one of ω-6 fatty acid desaturase is one, it is more important plant enzyme that catalyzes the formation of polyunsaturated fatty acids, linoleic acid catalysis oleic turn, determine the plant polyunsaturated fatty acid content and proportion. ω-3 fatty acids and ω-6 fatty acids belong to the polyunsaturated fatty acids, and the bodys own synthesis, must be from food intake, ω-6 / ω-3 fatty acid ratio is too high is not easy, less than five is appropriate. In this experiment oil was Peony Feng?Dan?Bai as the experimental material were taken ten days after flowering twenty days, thirty days peony seeds, the use of CTAB-LiCl total RNA was extracted peony seeds, then cDNA was synthesized . By semi-quantitative PCR was FAD2-3 peony gene expression during seed development in different variations, laid the foundation for further study of the function of the gene.
